Two 'extinct' trees rediscovered in Africa

Two tree species once feared extinct have been discovered in a threatened coastal forest in the African country of Tanzania. The two species have made reappearances before, but both trees, one a legume, the other a member of the mint family, were thought to have finally been obliterated in recent years.

Botantists rediscovered the trees in the southeastern African nation in 2011. The country has offered up another, perhaps more striking, discovery in recent months: a colorful new snake species.

"Both trees are still in critical danger of extinction, given that fewer than 50 individuals of each species are known," Roy Gereau, a botanist with the Missouri Botanical Garden's Africa and Madagascar Department, said in a statement.

The species, Erythrina schliebenii, a type of coral tree, has spectacular red flowers and vicious spines along its trunk. The tree was collected only twice, in the 1930s, from an area later cleared for a cashew plantation.

It was listed as "Extinct" on the IUCN Red List in 1998, but was rediscovered in a small patch of unprotected forest in 2001. When that forest was cleared for a biofuel plantation in 2008, it was again feared the thorny tree had gone extinct.

The other tree species, Karomia gigas, was known from only a single specimen cut down a few years after it was first discovered in coastal Kenya in 1977.

Another specimen was discovered in 1993 nearly 400 miles (600 kilometers) away in a tiny fragment of forest in Tanzania, but a more recent search at the same site didn't uncover any of the trees.

Last year, botanists from Tanzania's University of Dar es Salaam set out to look for both species, and discovered small populations of both trees in a remote coastal forest in southeastern Tanzania.

There are indications that recent improvements in infrastructure, together with a rapid population increase, are putting the coastal forests of the region under increasing threats from degradation and clearing.

    Genes for learning, remembering and forgetting

    ScienceDaily (Mar. 29, 2012) ? Certain genes and proteins that promote growth and development of embryos also play a surprising role in sending chemical signals that help adults learn, remember, forget and perhaps become addicted, University of Utah biologists have discovered.

    "We found that these molecules and signaling pathways [named Wnt] do not retire after development of the organism, but have a new and surprising role in the adult. They are called back to action to change the properties of the nervous system in response to experience," says biology Professor Andres Villu Maricq, senior author of the new study in the March 30 issue of the journal Cell.

    The study was performed in C. elegans -- the millimeter-long roundworm or nematode -- which has a nervous system that serves as a model for those of vertebrate animals, including humans.

    Because other Wnt pathways in worms are known to work in humans too, the researchers believe that Wnt genes, the Wnt proteins they produce and so-called "Wnt signaling" also are involved in human learning, memory and forgetting.

    "Almost certainly what we have discovered is going on in our brain as well," Maricq says. And because a worm nerve-signal "receptor" in the study is analogous to a human nicotine receptor involved in addiction, schizophrenia and some other mental disorders, some of the genes identified in the worm study "represent possible new targets for treatment of schizophrenia and perhaps addiction," he adds.

    Wnt genes and their proteins already were known to "pattern the development and distribution of organs in the body" during embryo development, and to be responsible for various cancers and developmental defects when mutated, he says.

    Maricq conducted the study with these Utah biologists: doctoral students Michael Jensen and Dane Maxfield; postdoctoral researchers Michael M. Francis, Frederic Hoerndli and Rui Wang; undergraduate Erica Johnson; Penelope Brockie, a research associate professor; and David M. Madsen, a senior research specialist.

    Synapse Plasticity is the Basis of Learning and Memory

    Synapses are the connections between nerve cells (neurons). Nerve signals are transmitted through synapses. Learning and memory concern how these connections are made, broken, strengthened or weakened. Proteins known as receptors are delivered to the synapses or removed from them to strengthen or weaken the connection.

    In the new study, Maricq and colleagues identified a "Wnt signaling pathway" -- a series of genes and the proteins they produce -- that controls the strength of nerve signal transmission from one neuron through a synapse to the next neuron. This allows "plasticity" of synapses -- a key factor in learning, retaining memories and forgetting.

    "The adult nervous system is not a stagnant tissue, but rather dynamic and plastic, with the strength of synapses -- specialized neuron-to-neuron connections -- changing with experience, learning and memory," Maricq says. "It's not a fixed thing, like when you're done making the heart, you're done."

    When synapses and thus incoming nerve signals are strengthened by adding receptors, an organism learns and remembers; when the opposite occurs, the organism forgets, he adds.

    How is that connection strengthened or weakened? When one neuron sends a nerve signal to another neuron, the first neuron releases a chemical known as a neurotransmitter, which moves through the synapse connecting the two cells and attaches or binds to receptors on the surface of the second neuron.

    "You can think of the receptors like amplifiers, like hearing aids," Maricq says.

    The volume of the received nerve signal depends on the number of receptors, which are stored in a supply depot just below the nerve cell's surface.

    The Wnt signaling identified in the new study "tells the depot to put more receptors into the synapse -- or not," says Maricq.

    He emphasizes that the Wnt chemical signal is different than the actual nerve signal carried by a neurotransmitter chemical, which in the new study was acetylcholine. The Wnt signal "is a secondary signal that controls the volume of the neurotransmitter signal," Maricq says.

    Worms Reveal Details of Nerve Signal Volume Control

    By crippling various genes in the worms, the researchers identified the "signaling pathway" by which a Wnt protein in one nerve cell sends a chemical signal to another cell telling it to increase the number of receptors on its surface, thus increasing the strength or volume of nerve signals between the cells.

    The type of nerve-signal receptor in the study is an acetylcholine receptor named ACR-16. When researchers crippled the gene that makes the ACR-16 receptor protein, there were not enough receptors, so nerve signals were disrupted and the worms "had uncoordinated movement," Maricq says. "They were semi-paralyzed."

    The scientists found mutations of other genes that also resulted in inadequate ACR-16 receptors and impaired the worms' movement. They discovered such genes belong to the "Wnt signaling pathway" that puts enough receptors on the cell surface so signals can be received.

    Besides ACR-16, genes in that pathway produced proteins named CWN-2 -- which is a Wnt protein -- LIN-17, CAM-1 and DSH-1.

    Here is how that pathway controls the volume of incoming nerve signals:

    1. A neuron releases CWN-2, which binds onto a receptor protein on the signal-receiving cell. That protein is a newly discovered combination of the LIN-17 and CAM-1 proteins.

    2. The LIN-17/CAM-1 protein sends a signal to a protein called disheveled, or DSH-1.

    3. "DSH-1 somehow sends the volume-control signal" that dispatches more ACR-16 receptors from depots inside the second neuron to that cell's surface, thus boosting the volume of the received nerve signal, Maricq says.

    The researchers used a green jellyfish protein to mark the ACR-16 receptors so they were visible under a microscope. When any of the genes in the Wnt signaling pathway were mutant, the scientists could see the green-labeled receptors accumulate under the surfaces of nerve cells instead of moving to the surface.

    Another experiment recorded electrical currents in worm nerve synapses and found it was smaller when any of the Wnt pathway genes were mutated. The smaller current -- reflecting impaired nerve-signal transmission -- explains why the mutant worms were partially paralyzed.

    Human Version of Worm Receptor Tied to Mental Disorders

    The ACR-16 acetylcholine receptor is the worm version of the alpha-7 nicotinic acetylcholine receptor in humans and other vertebrates. Both are similar in structure and function in animals from worms to fruit flies, mice and people.

    The alpha-7 receptor "is important in schizophrenia and a number of different mental disorders, and may have a role in addiction, but we don't understand how it's regulated," Maricq says.

    Many existing psychiatric drugs modify synapse strength. The new study suggests research should be done to show if the same Wnt signaling genes in worms also control alpha-7 receptor levels on human brain cells. If so, new drugs might be developed to target those genes as a way to treat mental disorders, including addiction.

    "Addiction is like learning at a primitive level," Maricq says. "Addiction means that somewhere in your brain, synapses are too strong. So you want more."

    The study was funded by the National Institutes of Health and the American Heart Association.

    This German-inspired brewpub is a definite gem in the Uptown district of Minneapolis. Located on the corner of Lake Street and Lyndale Avenue, The Herkimer has quite the selection of handcrafted bar fare and German beer made in house. As seen in the several brew kettles, The Herkimer takes pride on the local beer it creates. Celebrating their 10 Year Anniversary, The Herkimer pub is no stranger to awards. Boasting a Gold Medal at the Great American Beer Festival and securing the ?Best Fried? and ?Best Burger? awards from the City Pages, this brewery and pub is a staple establishment in Uptown.

    From the outside, The Herkimer Pub and Brewery looks like any other hole in the wall bar. From the street, it looks like any other dive bar on the block, but once inside, it?s a whole different story. The brew kettles are immediately in sight when first entering the eclectic bar. You know when they show off their own brewing devices they are damn sure of their product. The oversized, wraparound bar takes up most of the middle space in the area but seats are all around the restaurant as well. But, I was fortunate to get a prime stool by the taps so I could further study the pint I was going to get.

    The Herkimer offers 19 different varieties of beer on their menu. All ranging from light, crisp pints to dark, full-bodied brews, each offers a unique palate pleasing taste. The Dunkel Weiss was a strong pint that went down with ease. The cloudy, reddish color of the ale added to the smooth aroma of the hops. Having tasted several other beers, this wheat ale was by far my favorite. The Handy?s Lager was a simpler, light beer with a refreshing after taste. It was definitely a pint I could drink more than one of.

    A glance at the menu showed some very different choices than I was used to as normal bar food. Szechuan green beans, pub pretzels and fried pickles are just a few oddities listed in the menu. Seeing the sweet potato fries were voted ?Best Fries,? I had to try them. And, the award was justly given as the crispy, well-cooked fries paired perfectly with the zesty, delicious chipotle ketchup.

    The Herkimer has a very welcoming atmosphere with an array of patrons surrounding its bar. The bartenders are very knowledgeable when it comes to the beer brewed in the kettles and are more than happy to let you know which are the best pints. The Herkimer Pub and Brewery has happy hour seven days a week from 3 to 6 p.m., with 2-for-1?s on beer, well drinks and wine and discounted food. A late night happy hour is available from 10 p.m. to 1 a.m., with $3.50 beer, well drinks and wines and discounted food.

    Plant research reveals new role for gene silencing protein

    ScienceDaily (Mar. 29, 2012) ? A DICER protein, known to produce tiny RNAs in cells, also helps complete an important step in gene expression, according to research on Arabidopsis thaliana. The expression of a gene, when an organism's DNA is transcribed into a useable product, requires activation via a promoter or an external trigger. Plant research to be published in Science helps to show that later stages of transcription are just as important. This is likely to apply to other organisms, including humans.

    Termination is the final stage of transcription. Successful termination is dependent on DNA being transcribed into RNA with the correct sections, including a certain length tail.

    Scientists at the John Innes Centre on Norwich Research Park have found that where effective termination through the normal mechanisms has not occurred, DICER-LIKE 4 (DCL4) steps in to tidy up. Without termination, transcription continues down the chromosome unchecked.

    In this way, DCL4 plays a crucial and previously unknown role in transcription termination. It helps formation of the gene product. DCL4 is more commonly known to play a part in the opposite effect, gene silencing.

    "DCL4 is a back-up to termination processes, helping a gene to be successfully expressed," said lead author Professor Caroline Dean from JIC, which is strategically funded by BBSRC.

    The findings may help explain why gene silencing happens so often with transgenes. It was not known that so much attention should be given to the tail end of a gene.

    "Our research shows that for successful expression the end of a gene is just as important as its beginning," said Dean.

    When termination fails a lot of aberrant RNA is made -- this is degraded as part of a cell's quality control mechanism. This can have consequences for other sequences in the genome that match the aberrant RNA.

So, she ended up wasting thousands on advertising and promotions aimed at diners and families, which did attract some people, but very few returned. After 18 months in business, she ran out of money and then she ran out of places to borrow money.

    Interestingly, the pub was then bought by someone who transformed it into a very successful country restaurant, bar and seminar / meeting venue, with a reputation for great food and a warm, welcoming atmosphere. The place has a clear identity now, as a quality venue. It attracts people who are happy to pay a premium for quality. It doesn?t attract people looking for a cheap place to get drunk.

    We have to choose

    I wrote recently about how we need to match our fees or prices to our promises. Well, we also need to check that our business is balanced correctly, that people know?who we are and where we fit into the quality / price spectrum. For example, a designer with promotions all over their website for bargain $50 logo designs, is going to have trouble convincing us they are worth paying $5,000 for their premium design service. We know they do $50 work, that they work at the bottom end of the scale - The best designers don?t do that.

    Where do you pitch your prices or fees and what messages do you think that gives people? Are you trying to attract both, premium clients and budget conscious clients? If so, are you aware this usually makes you look like poor value, for the premium clients?

    Marketing Tip

    Take some time to review the type of client you are attracting (premium focused or budget conscious.) If you want more premium clients, but tend to attract the fee sensitive crowd, there?s ALWAYS a reason. Start by looking at your marketing messages and pay special attention to anything that suggests you serve the cheap end of the market.

    Just like the pub owner I mentioned earlier, we have a decision to make, if we want to attract the best clients and the most attractive fees.

    Titanoboa: How did a snake ever get so big?

    Titanoboa: The new Smithsonian exhibit in Grand Central Station displays a replica of the largest snake in history, the 48-foot titanoboa. Why don't huge snakes exist today??

    "Bigger. Badder. Boa," declares the banner of the swanky Smithsonian titanoboa exhibit in Grand Central Station, New York City. The replica of the ancient, and, thankfully for humans, extinct, snake measures 48 feet long. Its diameter is that of a manhole. The sinewy hulk currently lies on the cold marble of Grand Central, jaw-loosened, draconic head arcing back as it swallows a spiny crocodile, its hexagonal scales gleaming with venue lighting and overhead chandeliers.

    The replica of the 2,500-pound snake was unveiled today at the commuter hub to stimulate public interest in science, and promote the Smithsonian's more extensive titanoboa exhibit in D.C., which opens next week.

    Bones of the titanoboa were first discovered in a Colombian coal mine by a team of paleontologists led by Jonathan Bloch of University of Florida and Carlos Jaramillo of the Smithsonian Tropical Research Institute in Panama. They minted the snake's name, and published their discovery in 2009. Their discovery trumped that of the Eocene epoch's Gigantophis as the largest snake ever discovered.

    Comparison to modern snakes is even starker. The world's longest snake, the reticulate python, stretches a little more than half the length of the titanoboa. The green anaconda, the world's heaviest snake, is about a tenth of its weight.?

    Primeval hugeness isn't exclusive to snakes, either. Prehistoric animals seemed to have been much, much larger than they are today. The plant-eating Argentinosaurus is thought to have measured more than 100 feet long and weighed over 100 tons. The Quetzalcoatlus had a 45-foot wingspan. The water-bound Icththysaur was roughly 50 feet long, weighing 30 tons, a rough hybrid of a dolphin and a blue whale. The Ground Sloth was the size of today's elephant.

    What's with this trend? Do we know why prehistory was so redolent with animals that dwarf those we see today?

    Well, first, not all prehistoric animals were giant. Woolly mammoths aren't thought to have been significantly larger than today's elephant. The horse's earliest known ancestor is thought to have been the size of a house pet. And let's not?forget?that the largest animal ever, the krill-siphoning, 200-ton blue whale, is still with us today, although it is?endangered.

    Given those exceptions, though, prehistoric animals were markedly big, and scientists continue to ponder why. ?

    Large animals tend to be better predators and worse prey. After all, who is going to hunt and eat a titanoboa? Even non-predatory animals used their size to compete for resources, like plants.?Big animals tend to live longer and reproduce less often, which causes them to evolve more slowly than other species.?The perks of their size, however, made them particularly vulnerable to wide-scale extinction events. After such a cataclysm, it can take millions of years for huge animals to reappear.

    Body size may have also been a response to the changing environment. There's modern evidence that suggests animals are shrinking due to the warming planet. Inversely, when the ice ages occurred, warm-blooded animals increased in size to retain heat. Cold-blooded ones favored large bodies in warmer climates to better insulate them from overheating.

    Another theory suggests that size, particularly in dinosaurs, was fueled by rampant vegetation. Some 65 million years ago, in the Cretaceous period, atmospheric carbon dioxide was much more prevalent than it is today (note that this is not contradictory to anthropogenic global warming, which is operating on a much shorter timescale). Increasing carbon dioxide levels tend to drive up temperature, and so much of the Cretaceous world is thought to have been carpeted with thick vegetation. With this as their food base, dinosaurs weren't forced to shrink into forms that required less food, less energy, as there was plenty for them to eat.

    What about living things that aren't animals? The various scientific quests to discover the world's largest organism may not have to delve into prehistoric time. Even the blue whale doesn't compare to the 275-foot?General Sherman Sequoia, with a trunk that weighs over 2,000 tons. And then there is the?grove of genetically identical aspen trees in Fishlake National Forest ? considered by some scientists to count as a single organism ? which span 106 acres, and is estimated to weigh over 6,000 tons.

    Feds charge another ex-Carter's exec with fraud

    ATLANTA (AP) ? Another high-ranking executive of kids' clothing maker Carter's Inc. is facing fraud charges after federal prosecutors on Tuesday charged the former president of the Atlanta-based company with cooking the firm's books.

    Joseph Pacifico, 62, was charged with securities fraud, filing false financial statements and falsifying records of a public company while he was president of the company. Pacifico left Carter's in December 2009 after the company disclosed prosecutors were investigating its finances.

    The charges came as part of a new indictment against 57-year-old Joseph Elles, a former Carter's senior vice president who pleaded not guilty last year to more than 30 charges that he lied about the company's financial situation and then sold shares of its stock when the price was artificially bloated.

    Pacifico didn't immediately return calls seeking comment on Tuesday. Elles' defense attorney declined to comment, but he has said he was confident his client would be exonerated.

    The indictment claims Pacifico was aware by April 2009 that Elles gave rebates worth tens of millions of dollars to Kohl's Corp., the firm's largest customer, and then hid those discounts in financial statements. It said Pacifico lied to senior managers, signed false documents and told employees not to disclose the rebates to cover-up the fraud.

    As a result, the company's quarterly financial reports didn't reflect the cost of the discounts between November 2006 and July 2009.

    The reports helped buoy the company's stock price, and Elles earned $4.7 million from selling stock shared between 2004 and 2009, according to authorities. The share price then plummeted by more than 20 percent when the company announced it discovered the accounting issues in December 2009.

    Elles and Pacifico could both face decades in prison if found guilty of all the charges. A court hearing for Pacifico has not yet been set.

    U.S. Attorney Sally Quillian Yates said the charges were a sign that prosecutors were willing to follow the evidence "even to the top of the corporate ladder."

    "Shareholders expect and deserve far more from their corporate leaders," she said.
